P-selectin

P-selectin is also known as granule membrane protein 140 (GMP-140) and platelet activation dependent granule external membrane protein (PADGEM). P-selectin is expressed by circulating platelets and endothelial cells. It is not only a component of the membrane of the alpha and dense granules of platelets, but also a component of the membrane of the Weibel–Palade bodies of endothelial cells. P-selectin is a 140kDa adhesion molecule, the largest one in the selectin family. Selectin is a single-chain transmembrane glycoprotein that can be expressed on endothelial cells, leukocytes and platelets, and has similar properties to C-type lectins. The selectin family has similar domains, including C-type lectin domain, epidermal growth factor-like domain, short consensus repeats (the number of repeat units in P-selectin is 9), a transmembrane region and a cytoplasmic domain. Selectins can regulate leukocyte recruitment and play an important role in inflammation. After the stimulation of inflammatory mediators, the expression level of P-selectin on endothelial cells was rapidly increased. At the same time, P-selectin can also be used as a biomarker of platelet activation. P-selectin mediates platelet rolling in inflammation, and its expression on activated platelets contributes to the adhesion of leukocytes and the formation of platelet aggregates. P-selectin regulates the interaction between blood cells and endothelium, and its soluble form can be used as a plasma predictor of adverse cardiovascular events.
