Osteopontin and Cancers

OPN is a highly phosphorylated and glycosylated sialoprotein protein that belongs to the non-collagenous proteins family SIBLING (small integrin-binding ligand, N-linked glycoprotein). OPN is a versatile protein that is not only expressed in osteoclasts and osteoblasts, involved in bone remodeling. It is also an integral part of the immune system, secreted by neutrophils, NK cells, T cells and B cells, and can activate immune cells and regulate the immune system. OPN can communicate with cells through integrins, and can also participate in intracellular interactions through the interaction between its RGD sequence and CD44v 6/7. In addition, OPN is involved in cell differentiation and the regulation of cytokine production. OPN can activate downstream signaling pathways including protein kinase C (PKC) and phosphoatidylinositol 3-kinase (PI3K/Akt) via phospholipase C-γ (PLCγ), and OPN can also activate JNK1 (c-Jun N-terminal kinase 1) signaling pathways through nuclear factor-inducing kinase (NIK) and MEKK1 (mitogen-activated protein kinase kinase kinase1). OPN mediates the progression of various tumors, including breast cancer, prostate cancer, melanoma, etc. Increased OPN secretion is associated with the proliferation of tumor cells. On the other hand, OPN is closely related to the metastasis of tumor cells in diseases such as lung cancer and melanoma. In addition, OPN may also act on tumor angiogenesis and endothelial cell recruitment through activation with vascular endothelial growth factor (VEGF) and hypoxia-inducible factor 1 (HIF-1α). The key role of OPN in tumor progression may make it an effective biomarker and anti-cancer therapeutic target.
