IL-17 is a cytokine originally isolated from murine T-cell hybridomas. In the early stage, it was considered to be a subtype of the cytotoxic T lymphocyte-associated protein (CTLA) family, so it was called CTLA-8. It was later renamed IL-17. IL-17 has six homologous molecules, namely IL-17A, IL-17B, IL-17C, IL-17D, IL-17E and IL-17F. Among them, IL-17A and IL-17F have a high degree of homology and have similar functions. IL-17A can be produced by several types of immune cells including T helper 17 cell. The receptor family of IL-17 has 5 members, including IL-17RA, IL-17RB, IL-17RC, IL-17RD and IL-17RE. The 5 receptors have sequence homology, and their shared structures include the fibronectin III-like domain in the extracellular region and the SEF/IL-17R (SEFIR) domain in the intracellular region. IL-17RA is the shared receptor of IL-17 family cytokines, and IL-17RB, IL-17RC and IL-17RE are the specific receptors of IL-17E, IL-17A, IL-17F and IL-17C, respectively. IL-17RD is involved in the regulation of IL-17A signal transduction. IL-17A and IL-17F can induce pro-inflammatory gene expression alone or in conjunction with TNFα, IL-6, G-CSF, IL-1, CXCL, CCL20 and matrix metalloproteinases. IL-17 can also upregulate pro-inflammatory genes by activating NF-κB, MAPK and C/EBP cascades. In addition, IL-17 can activate the Jak-Stat and Jak-PI3 K pathways. The protein Act1 is also closely related to the functional pathway of IL-17. Act1 can serve as an anchoring platform at the proximal end of the receptor to initiate a TRAF6-dependent signaling pathway and a TRAF2-TRAF5-dependent cascade activated by IL-17.
