IL-10

IL-10 is also known as human cytokine synthesis inhibitory factor (CSIF), which is a pleiotropic, immunoregulatory cytokine encoded by the IL10 gene. IL-10 is mainly produced by B cells, macrophages, dendritic cells (DC), CD4+ and CD8+ T cells. It is a homodimer formed by the combination of two subunits each composed of 178 amino acids. During infection, IL-10 can affect the activity of many types of cells in the immune system. IL-10 can mediate its effects on T cells and natural killer (NK) cells functions through its direct effects on monocytes and macrophages. In addition, IL-10 can reduce the expression of MHC class II molecules on the surface of DCs and macrophages, weakening the function of antigen-presenting cells (APCs). It can also reduce the expression of proinflammatory cytokines and chemokines, thereby reducing the inflammatory response. IL-10 in CD4+ T cells can inhibit the production and proliferation of cytokines such as IFN-γ and TNF-α, so it can directly regulate Th1 and Th2 responses by inhibiting T cell activity and differentiation. IL-10 participates in the regulation of JAK-STAT signal transduction pathway through its receptor IL-10 receptor-1 and IL-10 receptor-2, thereby exerting its biological functions. IL-10 can protect the host from infection-related immunopathology. In patients with autoimmune diseases such as systemic lupus erythematosus (SLE) and multiple sclerosis (MS), IL-10 has abnormal serum levels, and the imbalance of IL-10 expression in patients may play an important role in these diseases.
