IFN-γ

IFNs are pleiotropic cytokines with anti-tumor and immunomodulatory properties. IFNs can be classified according to genetic loci, sequence identity, cell of origin, distribution of receptors, etc. IFN-α and IFN-β constitute the human type I IFN family, and IFN-γ is the only member of the type II IFN family. IFN-γ is mainly secreted by activated lymphocytes, but also by B cells, natural killer (NK) T cells and professional antigen-presenting cells. It is a homodimer formed by the non-covalent bonding of two polypeptide subunits, and has undergone multiple N-glycosylation during the synthesis process, and finally formed a 50kDa molecule. The biological effects of IFN-γ are achieved by activating molecular signaling pathways in cells. The combination of IFN-γ and its receptors IFNγR1 and IFNγR2 can activate the JAN/STAT pathway, thereby regulating the transcription of hundreds of genes and mediating different biological responses. The pleiotropic functions of IFN-γ are mediated by apoptosis and cell cycle regulators, inflammatory signaling molecules, transcriptional activators, etc. IFN-γ mainly activates anti-cancer immunity by promoting the activity of immune-related cells (including macrophages, NK cells, dendritic cells, etc.). It can also inhibit the differentiation and function of regulatory T cells, Th2 and Th17. IFN-γ is a major participant in human systemic autoimmune responses, and abnormal expression of IFN-γ can be found in autoimmune diseases. At the same time, its cellular targets may have the application prospect of prediction and treatment in tumor.
