Intercellular Adhesion Molecule-1

ICAM-1 is a transmembrane glycoprotein with a size of about 100kDa and belongs to the immunoglobulin supergene family. ICAM-1 is expressed in leukocyte subsets, endothelial cells, fibroblasts, glial cells, epithelial cells, etc. The full-length isoform of ICAM-1 consists of five Ig domains, a transmembrane domain, and a short cytoplasmic tail containing multiple threonine residues, which can interact with actin cytoskeleton and the ezrin/radixin/moesin complex associated with the lipid rafts. Membrane-bound ICAM-1 plays an important role in lymphocyte activation, leukocyte trafficking and some other immune functions. ICAM-1 can bind to a variety of ligands, including LFA-1, Mac-1, fibrinogen, hyaluronan and major rhinoviruses. The circulating form of ICAM-1 in body fluids, namely soluble ICAM-1 (sICAM-1), is produced by the proteolytic cleavage and alternative splicing of membrane-bound ICAM-1. sICAM-1 can also interact with LFA-1 and Mac-1. sICAM-1 is often studied as a biomarker and modulator that can promote inflammation. Its serum level is generally low, but it will increase in many inflammation-related pathological processes. Therefore, in a variety of diseases, ICAM-1 can be considered as a biomarker related to disease progression and severity.
