HER2 is one of the members of the human epidermal growth factor receptor (HER) family, which also includes HER1, HER3 and HER4. HER2 is a transmembrane protein with tyrosine kinase activity, composed of three parts: extracellular domain, transmembrane domain and intracellular tyrosine kinase active domain. HER2 was first discovered in the glioblastoma cell line of rats and was named Neu. Later, it was also known as ErbB2 because it was found to be structurally similar to avian erythroblastosis oncogene B. Finally, it was found that the protein encoded by this gene is similar to the structure of human epidermal growth factor receptor, so it is now often called HER2. HER2 gene is one of the important proto-oncogenes. As a transmembrane protein, HER2 can combine with growth factors on the cell surface to activate downstream signaling and promote cell survival, proliferation, differentiation and angiogenesis. HER2 can form a dimer, and two HER2 monomers or HER2 with other members of HER family to form a homodimer or heterodimer of HER2. The combination of dimer and ligand promotes protein phosphorylation and further activates downstream signaling pathways, including mitogen-activated protein kinase (MAPK), phospholipase C γ, phosphoinositide 3-kinase (PI3K/AKT), signal transducer and activator of transcription (STAT) and protein kinase C (PKC) signaling pathways. HER2 is a prognostic indicator for clinical treatment monitoring and an important target for the selection of tumor-targeted therapeutic drugs. Especially in the research and treatment of breast cancer, HER2 is an important biomarker.
