APRIL and Cancers

APRIL is one of the members of the tumor necrosis factor (TNF) superfamily, also known as tumor necrosis factor ligand superfamily member 13 (TNFSF13) and TRDL-1. APRIL has a high homology with B-cell activating factor (BAFF), which is another member of the TNF family. Both of them play an important role in the researches of autoimmune diseases and cancers. BAFF can bind to three receptors: B cell activating factor receptor (BAFF-R), B-cell maturation antigen (BCMA) and transmembrane activator and calcium modulator and cyclophilin ligand interactor (TACI), while APRIL binds to BCMA and TACI. BAFF and APRIL have important effects on various immune functions, including the maturation and differentiation of B cells and the production of antibodies. APRIL not only have abnormal expression in autoimmune diseases (the mRNA levels in patients are higher than those in the normal population), but also have abnormal expression in various tumor diseases. For example, in non-small cell lung cancer (NSCLC), the expression level of APRIL is up-regulated, and researchers believe that APRIL has potential as a diagnostic biomarker for NSCLC. In addition, APRIL is related to the growth and apoptosis of certain tumor cells. It has also been studied whether it can be used as an effective biomarker for other cancers, including gastric cancer, primary central nervous system lymphoma (PCNSL), colorectal adenocarcinoma, pancreatic adenocarcinoma, etc.
