Angiopoietin-2

Ang-2 is a highly active octapeptide produced by the cleavage of angiotensin I. Ang-2 is a typical cardiovascular mediator that plays an important role in regulating blood pressure and fluid homeostasis. Ang-2 receptors mainly exist on lymphocytes and macrophages, including AT1R and AT2R. A single AT1R is expressed in human cells, but there are two subtypes of AT1AR and AT1BR in mouse and rats, among which AT1AR has higher homology with human AT1R. AT1R consists of 359 amino acids and is an activator of the Gq/11 family. Ang-2 mainly induces its pro-stress and proinflammatory effects by stimulation of AT1R, so AT1R blockers have anti-inflammatory effects. AT2R is composed of 363 amino acids and can activate protein tyrosine phosphatase. AT2R may have anti-proliferation, anti-inflammatory and anti-fibrosis effects. Ang-2 acts as a regulator in innate immunity and may activate innate immunity and adaptive immunity. Natural killer cells may produce Ang-2 and deliver it to the site of inflammation. The interaction between Ang-2 and AT1R can promote the migration of neutrophils and stimulate the interaction between leukocytes and endothelial cells. Dendritic cells can express AT1R and AT2R, the activation of the cells can mediate the immune activity of Ang-2, and the combination of AT1R and Ang-2 can enhance the migration, maturation and antigen-presenting capabilities of dendritic cells.
