Introduction

7-Ketocholesterol is produced from cholesterol via the epimeric cholesterol 7-hydroperoxides (cholest-5-ene-3β-ol-7-hydroperoxide), which decomposes into the epimeric 7-hydroxycholesterols (cholest-5-ene-3β, 7-diol) and 7-ketocholesterol. Cholesterol is a monounsaturated component of the cell membrane that is involved in the permeability and fluidity. Due to the presence of a double bond (carbon 5) in cholesterol, various cholesterol oxidation products can be produced by different reaction mechanisms and pathways. Excessive intake of cholesterol oxidation products may be harmful to health because cholesterol oxidation products may have negative effects on the body, such as atherosclerosis, cytotoxicity, mutation and carcinogenesis, and may be involved in some chronic and degenerative diseases, as well as disorders of cellular function and lipid metabolism. 7-Ketocholesterol is the major oxidation product of cholesterol found in human atherosclerotic plaques and is more likely to cause atherosclerosis than cholesterol in some animal studies. 7-Ketocholesterol inhibits the rate-limiting enzyme cholesterol 7α-hydroxylase in bile acid biosynthesis and strongly inhibits the rate-limiting enzyme 3-hydroxy-3-methylglutaryl-coenzyme A reductase (HMG-CoA reductase) in cholesterol biosynthesis. Because it is easy to form and is the most representative hydroxysterol, 7-ketocholesterol is often used as a marker for cholesterol oxidation.
